Known hypersensitivity to tioconazole


Warnings/Precautions

Not effective when applied to the scalp; may interact with condoms and vaginal contraceptive diaphragms; avoid these products for 3 days following treatment


Adverse Reactions

1% to 10%: Genitourinary: Vulvar/vaginal burning

<1%: Vulvar itching, soreness, edema, or discharge; polyuria

Mechanism of Action

A 1-substituted imidazole derivative with a broad antifungal spectrum against a wide variety of dermatophytes and yeasts, usually at a concentration less than or equal to 6.25 mg/L; has been demonstrated to be at least as active in vitro as other imidazole antifungals. In vitro, tioconazole has been demonstrated 2-8 times as potent as miconazole against common dermal pathogens including Trichophyton mentagrophytes, T. rubrum, T. erinacei, T. tonsurans, Microsporum canis, Microsporum gypseum, and Candida albicans. Both agents appear to be similarly effective against Epidermophyton floccosum.


Pharmacodynamics/Kinetics

Absorption: Intravaginal: Following application small amounts of drug are absorbed systemically (25%) within 2-8 hours; therapeutic levels persist for 3-5 days after single dose

Half-life: 21-24 hours

Elimination: Urine and feces in approximate equal amounts


Usual Dosage

Adults: Vaginal: Insert 1 applicatorful in vagina, just prior to bedtime, as a single dose; therapy may extend to 7 days
