Warnings/Precautions

Rare epileptiform convulsions have been observed within the first few hours of administration, especially in patients with a history of CNS pathology


Adverse Reactions

>10%: Central nervous system: Dizziness, drowsiness, headache

<10%:

Central nervous system: Insomnia, malaise, hallucinations, behavior changes

Gastrointestinal: GI effects, orange/red discoloration of urine

Hepatic: Elevated LFTs

Dermatologic: Rash, urticaria, pruritus

Renal: Proteinuria


Drug Interactions

May be synergistic with praziquantel


Mechanism of Action

Not fully elucidated; causes worms to dislodge from their usual site of residence (mesenteric veins to the liver) by paralysis and contraction of musculature and subsequently phagocytized

Absorption: Oral: Well absorbed

Metabolism: Extensive in the GI tract via oxidation

Half-life: 1-2.5 hours

Time to peak: 1-3 hours

Elimination: <2% unchanged in urine; up to 75% of metabolites excreted in urine


Usual Dosage

Oral:

Adults: 12-15 mg/kg as a single dose

May interfere with spectrometric or color reaction urinalysis
